First with regard to the one reg entry, it would be real easy to do it with KiX using WriteValue().

Beware that it is possible to have proxy settings per machine. These would reside in HKLM. If you use the IEAK and AutoConfigProxy feature it would be even easier. You would set it on the server and the clients would update automatically.
